The largest group of transcription factors in higher eukaryotes are C2H2 proteins, which contain C2H2‐type zinc finger domains that specifically bind to DNA. Few well‐studied C2H2 proteins, however, demonstrate their key role in the control of gene expression and chromosome architecture. Here we review the features of the domain architecture of C2H2 proteins and the likely origin of C2H2 zinc fingers. A comprehensive investigation of proteomes for the presence of proteins with multiple clustered C2H2 domains has revealed a key difference (...) between groups of organisms. Unlike plants, transcription factors in metazoans contain clusters of C2H2 domains typically separated by a linker with the TGEKP consensus sequence. The average size of C2H2 clusters varies substantially, even between genomes of higher metazoans, and with a tendency to increase in combination with SCAN, and especially KRAB domains, reflecting the increasing complexity of gene regulatory networks. Broad‐complex, Tramtrack, and Bric‐à‐brac/poxvirus and zinc finger (BTB/POZ) is a conserved domain found in many eukaryotic proteins with diverse cellular functions. Recent studies revealed its importance in multiple developmental processes as well as in the onset and progression of oncological diseases. Most BTB domains can form multimers and selectively interact with non‐BTB proteins. Structural studies of BTB domains delineated the presence of different interfaces involved in various interactions mediated by BTBs and provided a basis for the specific inhibition of distinct (...) protein‐interaction interfaces. BTB domains originated early in eukaryotic evolution and progressively adapted their structural elements to perform distinct functions. In this review, we summarize and discuss the structural principles of protein‐protein interactions mediated by BTB domains based on the recently published structural data and advances in protein modeling. We propose an update to the structure‐based classification of BTB domain families and discuss their evolutionary interconnections. (shrink)

Pavel INCOMPLETE WORLDS, RITUAL EMOTIONS' IN recent years, the notion of "fictional world" has enjoyed a considerable rise in fortune. The expression, however, is not entirely new. To refer to the world of a literary work, of a novel or of a play, has always been a favorite way of speaking for literary critics and aestheticians. In most cases, these were informal worlds. A discussion of (...) the world of Balzac, Dostoyevsky, HenryJames, or Proust often amounts to a description of each author's preferences in die selection and rearrangement of his material. Such discussion primarily fits authors who purposefully attempt to build comprehensive representational artifacts. One speaks quite naturally about l'univers balzacien, or about the universe of Musil. But is one entitled to refer with equal ease to the world ofBenjamin Constant, meagerly perceptible in his Adolphe, a short novel built in the spirit of economy, displaying just a few characters and a minimum of worldly information? Perhaps density and comprehensiveness are not indispensable characteristics of literary worlds. Aestheticians and narratologists using the "fictional worlds" terminology seem to believe mat any kind of artistic text projects a world, be it rich or poor. Such worlds are imagined in the fashion of modern philosophy of logic as states of affairs, more or less comprehensive. Some writers believe that the states of affairs comprised in fictional worlds must be fairly sizable, since diey should include "not only what the author indicates, but also whatever is required (entailed) by that which he indicates." 1 The laws of nature which are not specifically contradicted by the text would belong to its world. Thus, a few notorious cases aside, there is no reason to doubt that every child born in fiction has been engendered by a human fadier as long as the text does not report an exception. And if it does? The article is devoted to identifying the concept of the meaning of life in the critical philosophy of A.I. Vvedensky, where special attention is paid to the methodological foundations and the historical and philosophical context. The formulation of the question about the meaning of life is one of the ultimate questions in philosophy, the answer to which makes it possible to determine the motives of human activity. In Vvedenskyʼs philosophy, the problem of goal-setting in life is revealed in the prism (...) of a key research intention – the construction of a scientifically revised worldview based on the principles of criticism. As a result, the question of life and its purpose is reduced to the formulation of the question of the logical permissibility and boundaries of questioning. The article attempts to reconstruct the philosopherʼs reasoning and identify the most significant definitions, which include morality, mental life, and the purpose of life. The disclosure of the concept of life in Vvedenskyʼs philosophy invariably requires consideration of the context in which the development of the philosopherʼs ideas takes place. Based on a comprehensive analysis, it is concluded that the concept of life is an extension of the main goal of Vvedenskyʼs research – the disclosure of the conditions for the formation of a reliable worldview, focused on the principles of scientific knowledge in accordance with moral values. Another important consequence is the provision on the priority of morality in relation to scientific and philosophical knowledge. The foundations of morality, to which the meaning of life belongs, can be presented only in a special form of applicability of critical reason – critical faith. Thus, the question of the specifics of critical metaphysics and the importance of differentiation of types and forms of faith in the work of the Russian neo-Kantian are clarified. (shrink)

This book explores the universe and its subsystems from the three lenses of evolutionary (diversifying), developmental (converging), and complex (adaptive) processes at all scales. It draws from prolific experts within the academic disciplines of complexity science, physical science, information and computer science, theoretical and evo-devo biology, cosmology, astrobiology, evolutionary theory, developmental theory, and philosophy. The chapters come from a Satellite Meeting, "Evolution, Development and Complexity" (EDC) hosted at the Conference on Complex Systems, in Cancun, 2017. The contributions will be peer-reviewed (...) and contributors from outside the conference will be invited to submit chapters to ensure full coverage of the topics. Investigation of the factors in natural history which conditioned the appearance of consciousness, that specifically human form of mental activity, necessarily presumes, in particular, a study of its functional preconditions or, in other words, of the higher forms of animal activity involving objects and of the corresponding mental processes. It is not enough to know the general psychological qualities of animals, the general principles by which their behavior is shaped, principles and properties offering evidence of a type of vital activity (...) qualitatively different from that of man. The traits of psychology and behavior, characteristic of the preponderance of the animal world, could not have become the immediate functional foundation capable of providing the real opportunity for the onset of the shaping of new regularities of mental activity. In the last 7 years, Russia has seen deep changes in all spheres of political and economic life. Some new realities have appeared in Russian medicine as well. This paper tells the story of how these changes came about, what kind of unusal situation were created, and how these situation are recognized in professional and public debates.

Insulators play a central role in subdividing the chromosome into a series of discrete topologically independent domains and in ensuring that enhancers and silencers contact their appropriate target genes. In this review we first discuss the general characteristics of insulator elements and their associated protein factors. A growing collection of insulator proteins have been identified including a family of proteins whose expression is developmentally regulated. We next consider several unexpected discoveries that require us to completely rethink how insulators function (and (...) how they can best be assayed). These discoveries also require a reevaluation of how insulators might restrict or orchestrate (by preventing or promoting) interactions between regulatory elements and their target genes. We conclude by connecting these new insights into the mechanisms of insulator action to dynamic changes in the three‐dimensional topology of the chromatin fiber and the generation of specific patterns of gene activity during development and differentiation. The paper is devoted to the analysis of theurgic aesthetics in relation to the concept of utopia that initiates a different understanding of the philosophy of the Russian diaspora representatives through the prism of utopian theurgy of beauty and art. Introducing the idea of utopian theurgy of beauty and art the authors emphasize its meaningful, axiological component. The authors interpret the utopian theurgy of beauty and art in the Russian diaspora philosophy of the first third of the 20th century as (...) an aesthetically mystical experience of the imperfection of human earthly existence that causes a gap with socio-historical reality with direct co-existence of the creative individual with God who is considered to be the true Beauty, the cosmic Harmony, and the universal Love. The meaning of the utopian theurgy of beauty and art is in achieving the goal of possible union with the deity, as well as in affirming persons actions as a co-creator of the world process. The utopian theurgy of beauty and art of an optimistic nature, associated with the light of hope directed to the transformation of earthly existence is reflected in N.S. Arsenyev's idea of spiritual joy achievable in the mystical experience of "longing for Beauty in God"; in B.P. Vysheslavtsev's reflections about the artistic and creative embodiment of eros as thirst, the birth of the God-Man in beauty, as well as in I.A. Ilyin's philosophy, in which, despite disappointments in social changes, the faith in art as the Mystery, as "a service and a joy" always following sacred traditions is preserved. The aesthetic and pessimistic foundations of utopian theurgy are realized in the aesthetic views of V.V. Weidle, according to which art has lost its orientation to high objective spirituality; in S.L. Frank's idea of unrealized Beauty as a dream of the ultimate transformation of the world that is opposed by the bitter reality of the split of being; as well as in the philosophy of N.A. Berdyaev, who postulates the tragedy of earthly existence caused by the unattainability of the full realization of the cosmic Beauty that considers as the goal of the world process. N.O. Lossky's metaphysics, intuitionism and value system are of particular importance, as the utopian theurgy of beauty and art consists in the following: there always remains a share of the absolute truth in art that is the world of illusions and artists imagination; the absolute value of beauty universally valid for all substantial personalities is affirmed. The article discusses the challenges, benefits, and risks that, from a bioethical perspective, arise because of the the development of eHealth projects. The conceptual framework of the research is based on H. Jonas’ principles of the ethics of responsibility and B.G. Yudin’s anthropological ideas on human beings as agents who constantly change their own boundaries in the “zone of phase transitions.” The article focuses on the events taking place in the zone of phase transitions between humans and machines in eHealth. (...) It is shown that for innovative practices related to digitalization and datafication in medicine, it is needed to rethink central bioethical concepts of personal autonomy and informed consent. In particular, the concept of broad or open informed consent is discussed, which allows the idea of moral responsibility in the field of biomedical technologies to be extended to events of uncertain future. The authors draw attention to the problems associated with the emergence of new autonomous subjects/agents in relationship between doctors and patients. The humanization of machines occurring in eHealth is accompanied by a counter trend – the formation of conceptions and practices of the quantified self. There emerges the practices of self-care and bio-power caused by the datafication and digitization of personality. The authors conclude that bioethics should proactively develop norms for the evolving interaction between doctor and patients. (shrink)

This essay poses the question whether it is possible, given the relationship of a subject and the method of its examination, at present as opposed to the time when Savigny worked, to talk about the same subject, and if it is not, what are the consequences for the interpretation of law. The turbulent history of Europe in the 20th century, marked by the cataclysms of world wars, genocides, expulsions and upheavals, therefore the necessity of new beginnings, jointly with other factors (...) (e.g. technological optimism) resulted not only in the ebbing of influence for religion, morality and tradition, but also in a deconstruction of law. This includes fragmentation, loss of the ability to formulate and interpret the purpose of law, as well as relativization of the traditional ordering principles of law (particularly the non-contradiction condition and contextual consistency, hierarchical structure, relative stability of law and its language). The opening of the texture of law, its deconstruction, creates a new segment of legislation, namely judicial law-making arising from the uncertainty of normative premises. (shrink)

Cílem této práce je připomenout odkaz J. B. Kozáka (1888–1974) a hodnotu jeho přednášek na Karlově univerzitě v letech 1949–1952, které měly hluboký význam v souvislosti s novou politickou situací po komunistickém puči v roce 1948. Život J. B. Kozáka byl hluboce ovlivněn jeho církevním vzděláním. Nakonec však získal post profesora filosofie na Karlově univerzitě v Praze. Jako prominentní zástupce Českého kulturního života se stal aktivním členem malého kroužku „pátečníků“ založeného spisovatelem K. Čapkem (1890–1938) a podporovaného T. G. Masarykem (1850–1937). (...) J. B. Kozák obhajoval demokratické ideje, ale jeho sympatie zároveň patřily také sociálně orientovaným levicovým osobnostem (viz například jeho přátelství s profesorem Z. Nejedlým, 1878–1962). Před válkou J. B. Kozák opustil Prahu a začal přednášet filosofii na Oberlin College v Chicagu (USA). Na konci války se však do Prahy vrátil. V létě roku 1945 získal znovu univerzitní post s možností přednášet. Ve vášnivých debatách s marxisty hájil myšlenky humanismu a svobody. Získal si sympatie svých studentů a stal se jejich morální autoritou, kterou zůstal i v těžkých časech politických čistek, kdy byli mnozí jeho kolegové propuštěni. Skutečnost, že si udržel pozici na univerzitě, nijak jeho autoritu nepoškodila. Byl chráněn kvůli svým levicovým názorům před válkou. Navzdory respektu, kterého se mu dostávalo, byla jeho možnost přednášet značně omezena. Připojeny jsou také úryvky jeho „dějin filosofie“ jako volné záznamy jeho přednášek. (shrink)

Introduction. The article is focused on analyzing the aesthetic aspect of A. Kojève’s philosophy, the ability of his philosophy, from an aesthetic point of view, to clarify a number of key problems of the modern political and cultural environment. The purpose of the study is to determine the epistemological attitude of A. Kojève’s philosophy able to clarify the way in which his philosophy problematizes the current cultural and political reality. Methods. Hermeneutics, comparative analysis and deconstruction are used as research methods. (...) Comparative analysis allows us to demarcate A. Kojève’s own line of reasoning, in the context of aesthetic and epistemological theories, as well as Hegelian themes themselves. Hermeneutics is the central method in the work, since the philosophy of A. Kojeve requires interpretation as an aesthetic theory. Deconstruction, formalized to the status of a method according to P. de Man, allows us to detect inconsistencies, the principle of mutual problematization of Kojeve’s philosophy and the current political and epistemological agenda. Scientific novelty of the research. The standard way of interpreting the work of A. Kojève is to evaluate him as an interpreter of G. Hegel. The author analyzes some meaning of A. Kojève’s philosophy, separate from G. Hegel’s explanation of the text. The novelty lies not only in the fact that A. Kojève’s philosophy is defined as theorizing that goes beyond technical hermeneutics (clear presentation), but also as a specific representation of such theorizing as epistemological aesthetic theory. The latter is a rather rare interpretation of the philosophy of A. Kojeve. This allows us to expand the traditional assumption that post-Hegelian epistemesthetics is the property of an extremely limited number of authors (for example, B. Croce). Moreover, such an interpretation of A. Kojève’s theories is a new element in the emerging trend of considering political issues within the framework of epistemological aesthetics. Results. The specific mode of interpreting G. Hegel, which is carried out by A. Kojève, turns the philosophy of the former into a unique aesthetic project. In this project, the category of aesthetic experience comes to the foreground, indicating the empty, formal nature of any discourse. In this regard, any claim to knowledge of reality, to a correct idea of the subject is replaced by the idea of creating the subject of discourse. Consistent treatment of this idea is symptomatic of the philosophy of A. Kojève. This philosophy is a reflection of those historical changes that gave rise to the culture of “aesthetic rationality”: empty and formal. In A. Kojève’s philosophy, one can see how such a culture of the End of History is, contrary to Hegelian ideas, not the era of the struggle decline, but the era of its strange transformation, a symptom of increasing political violence. Conclusions. A. Kojève’s philosophy is considered not as a utopian political project, but as a problematization of the modern cultural and political sphere. The latter is understood as an indication of the inconsistency of ontological discourses of Truth, the discovery in the very essence of discursive practices of an empty, formal principle, and therefore primarily of interest to aesthetics. The modern era of the End of History and the end of the era of persuasive speech gives an idea of the main political paradox (at least for the philosophy of A. Kojève, and at most for the modern political environment). The era of disappointment in discourse should become an era of refusal to fight for discourses, but it becomes an era of maximizing violence, since it is in a situation of disappointment that discourses can only be supported by force. We move from unconvincing speech to convincing violence, but it is possible to understand the origins and specifics of this transition only by considering A. Kojève’s philosophy in the light of such a discipline as aesthetics. (shrink)

The paper deals with the etymology of the personal names Qunï Säŋün and Toŋra Simä appearing in the ninth line of the Old Turkic Tonyukuk inscription. These names are borne by the envoys sent by the kagan of the Tokuz Oghuz to the Chinese and Khitan, respectively, to conclude a military alliance against the Turks. Both names have the same structure, a combination of an ethnonym vs. toŋra), referring to a tribal unit within the Tiele 鐵勒 confederation, and a title (...) of Chinese origin. Based on a thorough analysis of historical and philological data, the authors try to establish possible ethnolinguistic grounds for the Tokuz Oghuz mission, which have so far eluded researchers of the inscription and can be fully restored only in the light of the latest discoveries in the history and philology of Inner Asia of the Old Turkic period, e.g., the decipherment of the famous Khüis Tolgoi inscription from Mongolia. The study explores an approach to the philosophy of the sacred, tracing its presence from early recorded history to contemporary socio-political structures. The sacred is examined as a fundamental component of social organization, influencing social identities, institutions, and state legitimacy. -/- A key hypothesis introduced is the genetically determined centre of the sacred in the human brain, which evolved 50,000–70,000 years ago and functions archetypally, shaping emotions such as reverence, indignation at desecration, and numinous experiences. The sacred extends beyond traditional (...) religion into secular domains, including ideologies and eschatological narratives in modern societies. -/- The study also examines the role of trust and legitimacy in state stability, challenging N. Luhmann’s theory by arguing that trust operates beyond mere communication mechanisms and requires a fourth, sacred environment to maintain political order. The collapse of modern states (e.g., USSR, Libya, Syria) is linked to failures in reproducing trust. Furthermore, the paper introduces the concept of egregores – self-reproducing sacred structures that sustain collective belief systems, vital for the autopoiesis of political entities. -/- This work expands the philosophy of the sacred, linking it to macroevolutionary processes, the structuring of social identities, and the survival mechanisms of states. The boundary between analytic and synthetic sentences is well definable. Quine’s attempt to make it vague is based on a misunderstanding: instead of freeing semantics from shortcomings found, e.g. in Carnap’s work, Quine actually rejects semantics of natural language and replaces it by behavioristically articulated pragmatics. Semantics of natural language as a logical analysis is however possible and it can justify hard and fast lines between analyticity and syntheticity.

The author discusses B.G. Yudin's image technoscience as having two contours, the external one dealing with science, business and society, and the internal one represented by laboratories. Together these two contours present a multidimensional net of relations between science and technology in conducting experiments, development of instruments (e.g. visualization tools), etc. The author argues that, in such a system, coordinated activity of the internal and the external contours is provided by a synergy of regulatory principles of truth, good and usefulness. (...) He discusses a case of synthetic biology which, as he argues, demonstrates that the internal and external contours of technoscience undergo a process of technologization. Technologization is understood as a transition from research techniques to systematic use of technologies. Biotechnologies are claimed to have a double socio-biological helix network structure. Each network is formed as a kind of ad hoc "factory" and is aimed at solving specific research problems. (shrink)

The paper asks the question, whether nakedness embodies a potential wisdom. It deals with two different approaches to the phenomenon of nakedness: the first one rejecting, the second one appreciating the corporeality. The authors show different meanings the various cultures and civilizations attributed to nakedness, e.g. nakedness as a religious symbol of social subordination or belittling or of a specific national dominance . Attention is paid also to the meanings of nakedness in Jainism, where the nakedness is a symbol of (...) non-possessiveness; in Christianity, where the approach to nakedness is ambiguous, in tantrism with its vision of a person with numerous bodies, where the role of nakedness and sexuality is seen in cosmic context. Thus the meaning of nakedness remains multiple, depending on its context and visualization. The authors reject the pornography culture of contemporary society in favor of gymnosophy as the hierophany of the naturalness of nakedness. (shrink)

Open peer commentary on the article “Constructionism and Deconstructionism” by Pavel Boytchev. Upshot: Boytchev’s deconstructionism looks at first glance like a game of words. Upon a deeper view of the subject, he focuses our attention on the importance of deconstruction to the construction process, which is highly connected to creativity. In my contribution, I want to point out the close relationship of Boytchev’s deconstruction to the software development process, where requirements analysis corresponds to deconstruction and software design and implementation (...) correspond to construction. Book review on Pavel Gregorić – George Karamanolis (vyd.) Pseudo-Aristotle: De Mundo (On The Cosmos). A Commentary. Cambridge (Cambridge University Press) 2021, 245 str.

Extracellular nucleic acids are found in different biological fluids in the organism and in the environment: DNA is a ubiquitous component of the organic matter pool in the soil and in all marine and freshwater habitats. Data from recent studies strongly suggest that extracellular DNA and RNA play important biological roles in microbial communities and in higher organisms. DNA is an important component of bacterial biofilms and is involved in horizontal gene transfer. In recent years, the circulating extracellular nucleic acids (...) were shown to be associated with some diseases. Attempts are being made to develop noninvasive methods of early tumor diagnostics based on analysis of circulating DNA and RNA. Recent observations demonstrated the possibility of nucleic acids exchange between eukaryotic cells and extracellular space suggesting their participation in so far unidentified biological processes. The paper discusses the peculiarities of the analytic approach to the history of Ancient philosophy in the context of other, more popular approaches and genres. This approach is based on finding out an implicit argumentation and problems in the philosophical texts, and establishing logical connections between them. The paper also considers the perspectives of application of this approach to patristic texts. In addition, it shows the necessity of formalization and symbolization in the analytic history of philosophy.

A competing definition is the average distance measure proposed by Pavel Tichy and Graham Oddie. We will define three related, distance and size based, measures for actual truthlikeness and compare them with the two well-known options. However, we will start, Sect. 2, from a trio of such measures for nomic (...) truthlikeness. The nomic truth, or the true theory, here refers to what is nomically, e.g. physically, possible. In a nomic (and factual) context there are two basic kinds of theories, viz. either based on an exclusion claim or on an inclusion claim. Two-sided theories combine these claims, with the maximal claim as extreme special case. We will base truthlikeness measures for exclusion, inclusion, two-sided, and hence maximal, nomic theories on two similarity measures, one in terms of distances between conceptual possibilities and the other in terms of sizes of sets of such possibilities. In Sect. 3 we will treat actual truthlikeness as extreme special case of nomic truthlikeness, viz. assuming that there is just one nomic possibility, the actual one. Next we will compare the resulting measures mutually and with the above mentioned measures of Niiniluoto and Tichy & Oddie. Finally, in Sect. 4, we will sum up the results and explore five questions for further research. (shrink)
